The Protocadherin/Cadherin-related family is a group of calcium-dependent cell-adhesion proteins that play a crucial role in the establishment and maintenance of specific neuronal connections in the brain. Members of this family are involved in cells self-recognition and non-self discrimination, contributing to the sorting of heterogeneous cell types. They mediate homophilic trans-interactions through their cadherin domains, which bind calcium ions and facilitate adhesion. Some family members also participate in intermicrovillar adhesion, controlling the packing of microvilli at the apical membrane of epithelial cells and contributing to epithelial brush border differentiation. Additionally, certain protocadherins are implicated in synaptic reorganization underlying long-term memory and may be involved in regulating cell migration during developmental processes.
